Description
Proteinase K is a serine protease, which is a major kind of proteases produced by Tritirachiurn album Limber. Because microorganisms that can synthesize this protease can grow in an environment with kerantin as the sole carbon and nitrogen source, so it is named “proteinase K”. Proteinase K has extremely high enzymatic activity and broad substrate specificity. It can preferentially decompose ester bonds and peptide bonds adjacent to the C-terminus of hydrophobic amino acids, sulfur-containing amino acids, and aromatic amino acids. It is often used to degrade proteins to produce short peptides. It has the characteristics of the typical catalytic triplet Asp39-His69-Ser224 of serine proteases and has two Ca binding sites around the active center to increase its stability and maintain high enzymatic activity under a wider range of conditions. Proteinase K can inactivate or degrade proteins. Using this characteristic, proteinase K has important applications in the fields of nucleic acid purification, silk, medicine, food and brewing.
Currently commercially available proteinase K is mainly isolated and purified from Tritirachiurn album Limber by fermentation. As long as the medium contains protein, Tritirachiurn album Limber will be induced to produce proteinase K, so the fermentation medium generally needs to add BSA, milk powder or soybean meal as a nitrogen source. After the growth enters the stationary phase, the bacteria begin to secrete and express proteinase K. However, the biosynthesis of proteinase K during this process is inhibited by glucose and amino acids in the medium. In addition, the production of proteinase K by fermentation is difficult to scale up, so the production capacity of commercial proteinase K is generally relatively low.
