Proteinase K is commonly used in molecular biology to digest protein and remove contamination from preparations of nucleic acid. Addition of Proteinase K to nucleic acid preparations rapidly inactivates nucleases that might otherwise degrade the DNA or RNA during purification ideal for these applications because proteinase K is able to break down proteins and inactivate DNases and RNases that would otherwise degrade a desired sample of DNA or RNA.

Healthegn Biotech express and purify the recombinant proteinase K (rProK) from yeast (Pichia Pastoris) based on the optimized gene of Engyodontium album by site-directed mutation. The rProK is extensively purified to give highly active (more than 30U/mg, one unit of recombinant Proteinase K hydrolyzes urea-denaturated hemoglobin producing color equivalent of 1 μmol tyrosine per 1 min at 37°C and pH = 7.5) formulation free of any detectable nuclease activities. It is fully active up to 56°C and under denaturing conditions (e.g. in the presence of urea and/or SDS), what makes it ideal for genomic DNA extraction application. During the genomic DNA extraction, the rProK digest the protein associated with DNA, remove protein contaminants and rapidly inactivates DNase that might otherwise degrade the DNA. The rProK gives the best results in the presence of several other chemicals, such as Sodium dodecyl sulphate, EDTA, urea. In combination with these chemicals, rProK increases the efficiency of the result by increasing the purity of the DNA.

Using recombinant proteinase K (rProK) to extract genomic DNA, the Ct value was comparable with the commercial proteinase K, which meant the rProK could increase the efficiency of the result by increasing the purity of the DNA and was free of DNase.
