Description
Proteinase K is an important serine protease secreted by Tritirachium album Limber, containing the typical catalytic triad Asp39-His69-Ser224 characteristic of serine proteases, and has a strong ability to degrade natural proteins. It consists of 279 amino acids, containing 2 disulfide bonds and 1 free cysteine, which makes proteinase K maintain high stability in high temperature and extreme pH environment. Proteinase K contains two Ca2+ binding sites, which also increase the thermal stability as well as catalytic activity.
Proteinase K can be used for the digestion of a variety of proteins, including the preparation of chromosomal DNA for pulsed electrophoresis, western blotting, and the removal of nucleases in DNA and RNA preparations. The application of proteinase K in nucleic acid extraction involves many fields such as food-borne disease prevention, agricultural pest control, forensic gene identification and comparison, and molecular biology experimental research. Application objects include prokaryotes, human blood samples and tissues, animal and plant tissues, parasites, microorganisms, etc.
The currently commercially available proteinase K is mainly secreted by Tritirachium album Limber, which grows slowly and is difficult to cultivate at high density, and the yield of proteinase K is low. In addition, Tritirachium album Limber also secretes other proteases, which increases the difficulty of downstream separation and purification.
