Description
Proteinase K (EC 3.4.21.14) is a serine protease related to subtilisin, firstly was extracted and purified from Engyodontium album. It is named proteinase K because of its ability to degrade keratin. Proteinase K consists of a peptide chain containing 279 amino acid residues with a relative molecular mass of 28,930 Daltons. It is a highly active broad-spectrum protease, which can still maintain high enzymatic activity at 65°C and in the presence of EDTA, urea and detergents such as 0.5% sodium dodecyl sulfonate (SDS) solution or 1% Triton X-100, and has great advantages in extracting high molecular weight nucleic acids.
The separation and purification technology of nucleic acid is a basic technology of biochemistry and molecular biology. Nucleic acids are always combined with various proteins in cells. The separation of nucleic acids mainly refers to the separation of nucleic acids from biological macromolecules such as proteins, polysaccharides, and fats. When isolating nucleic acid, the contamination of other molecules should be excluded as much as possible to ensure the purity of the nucleic acid sample. Proteinase K has important applications in the separation and purification of nucleic acids, which can degrade proteins bound to nucleic acids and promote the separation of nucleic acids.
