Description
Proteinase K (EC 3.4.21.14) is a serine protease related to subtilisin, firstly was extracted and purified from Engyodontium album. It is named proteinase K because of its ability to degrade keratin. Proteinase K consists of a peptide chain containing 277 amino acid residues with a relative molecular mass of 28,930 Daltons. It is a highly active broad-spectrum protease, which can still maintain high enzymatic activity in denaturant urea or sodium dodecyl sulfonate (SDS) solution, so it is widely used for DNA or RNA extraction in various biological materials.
In the process of RNA extraction, proteinase K is always used for RNase inhibition and degradation. RNA quality is critical to the success of experiments such as Northern blotting and hybridization analysis, cDNA synthesis, and in vitro translation. RNA is extremely unstable and easily degraded by ubiquitous RNases, therefore, using proteinase K to create an RNase-free environment and strictly preventing RNase contamination is the key to successful RNA extraction, especially in Covid-19 virus RNA extraction.
Recombinant Proteinase K (rProK), derived from yeast, is purified by chromatography process with calcium and glycerin as protective agents without RNase and DNase. Compared to natural proteinase K, rProK has the same structure and function as natural proteinase K while it has higher purity greater than 95% and higher activity greater than 30 U/mg, as well as higher specific activity greater than 40U/mgP.
