Description
Proteinase K, a broad-spectrum serine protease, firstly was extracted and purified from Engyodontium album. Because it can degrade Keratin, so named proteinase K. The crystallization and molecular structure of Proteinase K indicate it belongs to subtilisin family, whose active site has a characteristic catalytic triple amino acid Asp39-His69-Ser224. Proteinase K has no significant substrate specificity, and main cleavage site is the peptide bond at the carboxyl end of the hydrophobic amino acid such as aliphatic or aromatic. It has extremely high enzyme activity and wide substrate specificity, which can remove the activity of RNase and DNase, and is usually used to degrade protein in various molecular biology and cell biology related experiments. In addition, it is also widely used in the detection of Novel Coronavirus (Covid-19) in In Vitro Diagnosis (IVD).
In the detection of the Covid-19, there is a high risk of infection in the pre-processing, extraction, and amplification of the nucleic acid amplification test. Therefore, it is necessary to inactivate the virus before the amplification and this requires the use of the virus sampling solution which is one member of the new coronavirus nucleic acid reagent group, while proteinase K is a very important component in the virus sampling solution. Virus sampling solution has two main functions which are both inseparable from the protein digestion ability of proteinase K, one is to inactivate the virus, denature the virus protein, and make it inactive, "dead" and infectivity, and then finally improve the safety of the transportation and detection stage; the second function is to crack the virus to release the nucleic acid and eliminate the nucleic acid decomposing enzyme (RNase) to prevent the virus from degrading the RNA.
Recombinant Proteinase K (rProK), derived from yeast, is purified by chromatography process with calcium and glycerin as protective agents. rProK is produced by Healthgen Biotech with its core patented protein purification platform and has high purity greater than 95% and high specific activity greater than 40U/mgP, so it can effectively improve the quality of proteinase K monoclonal antibodies in the preparation of it and then finally ensure the accurate test results in the content detection process of proteinase K.
