Greater Than 40 U/MgP Specific Activity Recombinant Proteinase K For DNA Extraction

Natural proteinase K is a powerful proteolytic enzyme purified from Tritirachium album limber. It has high activity and can operate in a wide pH range (4~12.5) and high temperature (50~70°C), and chelating agents such as EDTA or detergents such as SDS cannot inactivate it. Proteinase K is a key reagent for DNA extraction. In DNA extraction, the proteinase K’s main role is to enzymatically digest histones bound to nucleic acids to free the DNA in the solution, and then use different methods to extract, remove impurities, and collect the DNA.

Proteinase K has two Ca²⁺ binding sites with a certain distance from the active center of the enzyme, which is not directly related to the catalytic mechanism. However, if Ca²⁺ is removed from the enzyme, the catalytic activity of proteinase K will be lost by about 80% due to remote structural changes.
