Description
Human serum albumin (HSA) is the most abundant protein in plasma synthesized by hepatic parenchymal cells in human body, accounting for about 40%–60% of total protein in plasma. HSA exists in human blood with a concentration of 35–50 g/L and a long half-life of 19 days. HSA consists of 585 amino acids with a molecular weight of 66.5 KDa. Besides a huge amount of glutamic acid, lysine and arginine, 35 cysteine residues form 17 disulfide bonds to stabilize structure and retain physical and chemical properties of HSA in pH range of 4–6. In structure, HSA is composed of three homologous domains I, II, and III. HSA is largely responsible for maintaining, normal osmolarity in the bloodstream but also functions as a carrier for numerous small molecules. Clinically, HSA is used to treat severe hypoalbuminemia and traumatic shock, with usual dosage in excess of 10 g/dose.
To date, HSA has been produced by fractionation of human plasma. As the source of the blood can vary, there is the potential risk of HSA contamination by blood-derived pathogens. Human plasma is also in limited supply, recombinant human serum albumin becomes an alternative industrial production which greatly compensates for the shortage of HSA supply.
OsrHSA, a recombinant human serum albumin expressed by rice endosperm platform, provides an excellent solution for animal free material. Compared with the plasma HSA, rHSA has the same amino acids sequence, molecular weight, N- and C terminus, PI and crystal structure as plasma HSA.
