Description
Human serum albumin (HSA) is the most abundant protein component in human blood with a concentration of 35–50 g/L and a long half-life of 19 days, which plays a vital role in regulating the plasma osmotic pressure and the fluid distribution between compartments. HSA consists of 585 amino acids with a molecular weight of 66,439 Da. In structure, HSA is mainly composed of α-helices and forms multiple binding sites, which endows HSA as a carrier in the blood. These special domains form natural predominant drug binding pockets. In addition, one free thiol at Cys34 and seven fatty acid binding sites lay the foundation for the function of HSA as a carrier.
Because of these unique physiological functions of HSA, the HSA is utilized as a drug carrier in vitro, such as HSA-based anti-cancer drugs, due to its prominent advantages of good biocompatibility, favorable stability, high drug delivery capacity, targeting capability and a long half-life. Intensive investigations indicate that HSA-based therapeutics have shown a great potential in cancer therapy.
Currently, HSA is mostly derived from blood collected from multiple donors, which means pharmaceutical manufactures will suffer potential reduced source of HSA because of an interruption in the supply and potential existence of various infectious disease pathogens in the human blood, especially AIDS, hepatitis B and other viruses.
Healthgen Biotech recombinant human serum albumin(OsrHSA) was expressed in the rice endosperm and stored in the rice grain protein body which can keep the protein stability and bioactivity to the maximum extent. And the OsrHSA has been demonstrated to be folded into the identical structure as pHSA. Moreover, all fatty acid binding sites in OsrHSA were identified at the same locations as in plasma HSA. And two major drug binding sites found in pHSA were also present in OsrHSA .
The binding capacity assay of OsrHSA showed that the affinities of warfarin(site I) and naproxen(site II), two site-specific drug markers of HSA, is similar to the binding affinities observed to pHSA, indicating that no significant difference in drug-binding affinity exists between OsrHSA and pHSA.
