Engyodontium album Proteinase K Recombinant high purity
Description
Proteinase K, a broad-spectrum serine protease, firstly was extracted and purified from Engyodontium album. Because it can degrade Keratin, so named proteinase K. The crystallization and molecular structure of Proteinase K indicate it belongs to subtilisin family, whose active site has a characteristic catalytic triple amino acid Asp39-His69-Ser224. Proteinase K has no significant substrate specificity, and it cleaves peptide bonds adjacent to the carboxylic group of aliphatic and aromatic amino acids and is useful for general digestion of protein in biological samples.
Healthegn Biotech express and purify the recombinant proteinase K (rProK) from yeast (Pichia Pastoris) based on the optimized gene of Engyodontium album by site-directed mutation. The rProK possesses a high specific activity which remains stable over a wide range of temperatures and pH values with substantially increased activity at higher temperature. The stability of rProK in urea and SDS and its ability to digest native proteins make it useful for a variety of applications, including preparation of chromosomal DNA for pulsed-field gel electrophoresis, protein fingerprinting and removal of nucleases from preparations of DNA and RNA. A typical working concentration for Proteinase K is 50–100µg/ml.
