Description
Proteinase K is a highly active 29kDa serine protease isolated from the fungus Tritirachium album. It exhibits broad cleavage specificity on native and denatured proteins and is widely used in the purification of DNA and RNA. Its activity is increased in the presence of denaturants such as SDS and elevated temperature (50–60°C). Proteinase K is a broad-spectrum serine protease under the subtilisin-like class (there are two types of serine proteases, chymotrypsin-like and subtilisin-like). It belongs to Peptidase family S8. The predominant site of cleavage is the peptide bond adjacent to the carboxyl group of aliphatic and aromatic amino acids with blocked alpha amino groups.
Healthgen Biotech recombinant Proteinase K is extensively purified to give highly active formulation free of any detectable nuclease activities. It is fully active up to 56°C and under denaturing conditions (e.g. in the presence of urea and/or SDS), what makes it ideal for DNA extraction application. The recommended working concentration is 50–100 µg/ml for protein removal and enzyme inactivation, and up to 2 mg/ml for tissue treatment. The recombinant Proteinase K (rProK), lyophilized powder can be prepared as a 20 mg/ml stock solution in water. The rProK products are free of detectable DNase and RNase.
The rProK possesses a high specific activity which remains stable over a wide range of temperatures and pH values with substantially increased activity at higher temperature. Purified by chromatography process, the rProK has a high purity more than 95%. The rProK can be used in a variety of molecular biology, cell biology and other related experiments, such as genomic DNA extraction, enzyme digestion and removal, cell permeability and so on.
