Description

 

Proteinase K is a serine protease of the subtilisin type with strong proteolytic activity. The ‘K’ in the name indicates the digestion of keratine to cover the carbon/nitrogen requirements of the fungus. Proteinase K is an extremely nonspecific enzyme with a wide variability of cut peptide bonds. Proteinase K has two binding sites for Ca2+ ions which do not really participate in the catalysis, but do contribute to the structural stability of the protein. Due to the high ability of proteinase K to cut native proteins, Proteinase K is used for the destruction of proteins in cell lysates (tissue, cell culture cells) and for the release of nucleic acids, since it very effectively inactivates DNases and RNases. Some examples for applications: Proteinase K is very useful in the isolation of highly native, undamaged DNAs or RNAs, since most microbial or mammalian DNases and RNases are rapidly removed by the proteinase K.

 

Healthegn Biotech express and purify the recombinant proteinase K (rProK) from yeast (Pichia Pastoris) based on the optimized gene of Engyodontium album by site-directed mutation. The rProK possesses a high specific activity which remains stable over a wide range of temperatures and pH values with substantially increased activity at higher temperature. Purified by chromatography process, the rProK has a high purity more than 95%, which is also completely free of DNase and RNase activities.
