Description
Proteinase K is a broad-spectrum serine protease, and has no significant substrate specificity, and main cleavage site is the peptide bond at the carboxyl end of the hydrophobic amino acid such as aliphatic or aromatic. It plays a central role in the present method and is responsible for digestion of all the proteins in the cell lysate.
The protein is made up of the long chain of amino acids. Depending upon their nature of interaction with water, it is categorised into the hydrophilic (attract water ) and hydrophobic (water repel) amino acids. The proteinase K digests mainly the hydrophobic amino acids ( both aromatic as well as aliphatic). During the DNA extraction, the proteinase K digest the protein associated with DNA, remove protein contaminants and rapidly inactivates DNase that might otherwise degrade the DNA. The proteinase K gives the best results in the presence of several other chemicals, such as Sodium dodecyl sulphate, EDTA, urea. In combination with these chemicals, proteinase K increases the efficiency of the result by increasing the purity of the DNA.
Healthgen Biotech recombinant Proteinase K is extensively purified to give highly active formulation free of any detectable nuclease activities. It is fully active up to 56°C and under denaturing conditions (e.g. in the presence of urea and/or SDS), what makes it ideal for DNA extraction application.
