High bioactivity proteinase K for DNA and RNA purification
Description
Proteinase K is a typical member of the subtilisin family of proteinases. It consists of 278 amino acids, with molecular mass 29kDa, and the pI is 8.9. Proteinase K is an endopeptidase and carries at its active site the catalytic triad Asp39, His69, Ser224. Its full enzymatic activated by calcium, the enzyme digests proteins preferentially after hydrophobic amino acids. Although calcium ions do not affect the enzyme activity, they do contribute to its stability. Proteins will be completely digested if the incubation time is long and the protease concentration high enough. Proteinase K is also stable over a wide pH range (4–12), with a pH optimum of pH 8.0. An elevation of the reaction temperature from 37 °C to 50–60 °C may increase the activity several times, like the addition of 0.5–1% (SDS) or Guanidinium chloride, Guanidinium thiocyanate and urea.
Recombinant Proteinase K (rProK) is derived from yeast based on the optimized gene of Engyodontium album by site-directed mutation. It is purified by chromatography process. The rProK is completely free of DNase and RNase activities. The biological activity is more than 30U/mg, which one unit is defined as the amount of enzyme that will hydrolyze urea-denatured hemoglobin to produce color equivalent to 1.0 µmol tyrosine per min at 37°C, pH 7.5 (color by Folin-Ciocalteu reagent).
During the DNA and RNA purification, there is a lot of contaminating proteins present. These contaminants must be removed. Proteinase K, which is a broad spectrum serine protease, is used in many DNA and RNA purification protocols to digest these contaminating proteins. Addition of Proteinase K to nucleic acid preparations rapidly inactivates nucleases which may otherwise degrade the DNA or RNA during purification.
