Fibronectin (FN) is a ubiquitous extracellular matrix (ECM) protein that has been implicated in many essential cell processes including wound healing and embryonic morphogenesis as well as regulation of cell behaviors such as adhesion and migration. FN is a very large molecule, binding to other ECM structures, various proteoglycans, and growth factors (GFs), and many of these binding partners have the ability to direct cell behavior. FN is a dimeric adhesive glycoprotein composed of two approximately 220 kDa subunits connected by a pair of disulfide bonds at their C-terminal.

Fibronectin plays a major role in cell adhesion, growth, migration, and differentiation, and it is important for processes such as wound healing and embryonic development. Altered fibronectin expression, degradation, and organization have been associated with a number of pathologies, including cancer and fibrosis.

Currently, the FN is purified from the plasma, which however is limited by the availability of supply. In addition, the use of even small amounts of human- or animal derived proteins in serum-free medium is coming under increasing scrutiny because of disease causing agents in both human- and bovine-derived materials. In addition, use of non-serum media allows for easier purification. As the recombinant human fibronectin (OsrhFN) was produced in rice endosperm using chemically defined raw materials, the absence of animal- and human-derived viruses and other related contaminants is assured and would allow for cell culture media systems to utilise a safer, more defined and consistent raw material. The OsrhFN added in the xeno free medium for CHO cell culture, and the result showed the OsrhFN can promote cell growth.

The rice endosperm expression platform advantages are over other systems in terms of scalability, safety, and cost-effectiveness. In addition, the rice endosperm provides the ideal place in which the seed is not only providing a stable site for the deposition of recombinant proteins to prevent from the degradation by proteases, but also storing at ambient temperatures for several years.
