Fibronectin (FN) is a macromolecular glycoprotein with a monomer molecular weight of 220-250KD and a dimer molecular weight of about 450KD. There are three main types of fibronectins. The first type is plasma fibronectin (pFN), synthesized by hepatocytes or endothelial cells and exists in plasma and body fluids in a soluble dimer state; The second type is cellular fibronectin (cFN), and synthesized by types of cells, including fibroblasts, astrocytes, early mesenchymal cells, etc. cFN are deposited on the cell surface, extracellular matrix, intercellular matrix, basement membrane and connective tissue in the form of dimers or multimers; pFN and cFN are similar in molecule, structure and function, and can be transformed into each other; The third type is fetal fibronectin (fFN), mainly synthesized by embryonic tissue, placental tissue and malignant cells, etc., and produced by overexpression of the gene sequence EDA and EDB.

FN has a variety of biological activities. Modern medical researches show that the change of fibronectin content is closely related to various clinical diseases, such as rheumatoid arthritis, and the detection of fibronectin content has very important clinical significance in patients with rheumatoid arthritis.

Studies have shown that patients with rheumatoid arthritis may have accelerated biosynthesis of cellular cFN in or around the lesion due to the need for repair after tissue injury, and some of the cFN enters the blood, resulting in a secondary content increase of cFN in the plasma, so the concentration change of plasma cFN may be used as an important indicator of curative effect observation and prognosis judgment.

At present, the common fibronectin which are used to prepare FN polyclonal antibody in the market is mainly derived from human plasma or animal plasma as a subsidiary product of blood products, however, the output of blood derived FN is low and the supply is not stable due to the limited blood sources. There are relatively few manufacturers of recombinant human fibronectin, not only the production is low and the price is high, but most of them are 40-50KD recombinant human fibronectin small molecule fragments.
