Recombinant fibronectin for stem cell culture
Description
Fibronectin, an abundant and ubiquitous protein of the extracellular matrix, forms a fibrillar network by interacting directly with cell surface receptors. It is synthesized by a variety of different cells and is secreted as a disulfide-bonded dimer with 220 kDa subunits. FN is a mosaic protein composed of three types of repeating modular consensus amino acid sequences. Functional domains of FN mediate interactions with cell surface receptors and a number of other macromolecules including fibrin, fibrinogen, collagen, heparin, immune complexes, and fibronectin itself. So FN is known to play a key role in cell adhesion, spreading, migration, differentiation and proliferation via interactions with transmembrane integrin receptors.
Stemcells are defined by three essential features, as they are i) undifferentiated cells able to give rise both to ii) differentiated daughter cells and iii) cells that retain their stemness by self-renewal. The FN can be an player in stem cell culture, since it can directly or indirectly modulate the maintenance, proliferation, self-renewal and differentiation of stem cells, which is a key component of stem cell niches and is involved in various aspects of stem cell behavior, thus having a major impact on tissue homeostasis and regeneration under physiological and pathological conditions. Numerous studies has demonstrated that FN works very important role in the stem cell culture, like FN result in murine HSCs expansion of primitive stem cells and improvement in the marrow engraftibility.
Currently, the FN is purified from the plasma, which however is limited by the availability of supply. In addition, due to shortage of plasma and low expression levels of recombinant fibronectin in conventional gene expression systems, Healthgen Biotech is focusing on developing, manufacturing and marketing a series of environmental friendly and safe products by using the State-of-the-Art platform called OryzHiExp, having high capacity to express proteins or small peptides in the rice grain.
