Description
Fibronectin (FN) is a particularly important and well-studied component of the extracellular matrix, and is known to play a key role in cell adhesion, spreading, migration, differentiation and proliferation via interactions with transmembrane integrin receptors. FN is a 220 kDa glycoprotein composed of 2 subunits bound via a disulfide bond, and exists as a soluble circulating protein in body fluids like plasma, but can be rearranged to a fibrillar insoluble component in the extracellular matrix of cells. FN possesses anacidic isoelectric point (pI = 5.5–6.0), and so is negatively charged at a physiological pH (7.4).
FN molecule contains several functionally and structurally distinct domains which may bind to cell surfaces, collagen, fibrinogen or fibrin, complement, glycosaminoglycans, proteoglycans and heparin. FN can enhance cell adhesion and spreading and affect the routes of cell migration in cell culture, which is quite of importance for the cell growth and proliferation. FN is suitable as a thin coating on cell/tissue culture surface for a variety of cell types, which can also be used as an additive to serum –free culture medium.
