Recombinant fibronectin for coating agent in cell culture

 

Description

 

Fibronectin (FN) is an extracellular matrix glycoprotein that binds to transmembrane proteins called integrins. It is an important regulator in extracellular matrix remodelling. It is composed of two protein heterodimers linked by a pair of disulfi de bonds and exists in two major forms; cellular fi bronectin (an insoluble form of fibrils) and plasma fibronectin (a soluble form produced by hepatocytes in the liver). It contains several functionally and structurally distinct domains which may bind to cell surfaces, collagen, fibrinogen or fibrin, complement, glycosaminoglycans, proteoglycans and heparin.

 

Numerous studies have shown that FN may enhance cell adhesion and spreading and affect the routes of cell migration in vitro cell culture. So the FN is used as a coating reagent on the cell/tissue culture surface to promote attachment, spreading and proliferation of a variety of cell types, including endothelial cells, fibroblast cells and stem cells.

 

Currently, the FN is purified from the plasma, which however is limited by the availability of supply. In addition, due to shortage of plasma and low expression levels of recombinant fibronectin in conventional gene expression systems, Healthgen Biotech used the rice endosperm expression platform to express the recombinant human fibronectin (OsrhFN), with the world's top purification processing. The recombinant fibronectin (OsrhFN) has been demonstrated that has the same physical and chemical with the plasma derived FN.

The OsrhFN is suitable for cell culture coating reagent in vitro, including plastic ware, glassware, and microcarrier beads, and can also be used with variety of media including DMEM/F-12, basal media and supplement media. The OsrhFN binds the cell and cultureware surface to improve the cell adhesion, cell motility, opsonization, and maintenance of cell shape.

 

The use of even small amounts of human- or animal derived proteins in serum-free medium is coming under increasing scrutiny because of disease causing agents in both human- and animal-derived materials. In addition, use of non-serum media allows for easier downstream purification. As the OsrhFN was produced in rice endosperm using chemically defined raw materials, the absence of animal- and human-derived viruses and other related contaminants is assured and would allow for cell culture media systems to utilise a safer, more defined and consistent raw material.
