Description
Proteinase K from Tritirachium album is a subtilisin-related serine protease. It is an endopeptidase with a very high specific activity and a broad spectrum of action. It is very active in the presence of agents that denature proteins (e.g., SDS and urea). It destructs proteins in cell lysates and promotes
the isolation of nucleic acids and is therefore commonly used for nucleic acid preparation as it inactivates rapidly nucleases, such as DNases and RNase.
Recombinant Proteinase K (rProK) is derived from yeast based on the optimized gene of Engyodontium album by site-directed mutation. It is purified by chromatography process. The rProK is completely free of DNase and RNase activities. The biological activity is more than 30U/mg, which one unit is defined as the amount of enzyme that will hydrolyze urea-denatured hemoglobin to produce color equivalent to 1.0 µmol tyrosine per min at 37°C, pH 7.5 (color by Folin-Ciocalteu reagent).
During the nucleic acids extraction, the recombinant Proteinase K (rProK) is used in many DNA and RNA purification protocols to digest these contaminating proteins. Addition of rProK to nucleic acid preparations rapidly inactivates nucleases which may otherwise degrade the DNA or RNA during extraction.
